چکیده

Suppression subtractive hybridization was used to analyze the differences in gene expression in the body wall of the Japanese sea cucumber Apostichopus japonicus, induced by heat stress (28°C), in order to understand the response at the molecular level. A total of 737 clones was selected and analyzed, and 165 distinct sequences were obtained, with 65 sharing similarity to sequences in public databases. The sequenced genes were classified into 3 functional categories. Increased expression of some genes in response to stress was concomitant with decreased expression of genes concerned with energy production and metabolism. The results (1) suggest that heat stress has significant effects on gene expression in A. japonicus, and (2) provide the basis for further study of molecular regulation mechanisms underlying the response to heat stress and estivation in A. japonicus.

Understanding the Heat Shock Response in the Sea Cucumber Apostichopus japonicus, Using iTRAQ-Based Proteomics.

The sea cucumber Apostichopus japonicus is exploited as a commercial species owing to their high nutritive and medicinal value. Recent high summer temperatures have caused high mortality rates in A. japonicus. In this study, we applied the isobaric tag for relative and absolute quantitation (iTRAQ) technique to investigate the global protein expression profile under an acute short-term (48 h) h...

The Preventative Effect of Dietary Apostichopus japonicus on Intestinal Microflora Dysregulation in Immunosuppressive Mice Induced by Cyclophosphamide

Sea cucumbers are recognized as food and drug resources with many nutritional benefits, and Apostichopus japonicus is a kind of sea cucumber with good quality. Processing methods have some effect on its quality. This study aimed to explore the effects of Apostichopus japonicus with three different processing methods (dried, instant, and enzymatic sea cucummbers) on intestinal microflora dysregu...

Cloning and expression analysis of Wnt6 and Hox6 during intestinal regeneration in the sea cucumber Apostichopus japonicus.

Many developmental genes play important roles in regeneration. Here, we cloned the two developmental genes, Wnt6 and Hox6, from Apostichopus japonicus by rapid amplification of cDNA ends (RACE) polymerase chain reaction (PCR). The Wnt6 full-length cDNA is 1140 bp long with a 1029-bp open reading frame encoding 342 amino acids. The deduced Wnt6 protein contained a 22-amino acid signal peptide an...

Identification and Characterization of MicroRNAs from Longitudinal Muscle and Respiratory Tree in Sea Cucumber (Apostichopus japonicus) Using High-Throughput Sequencing

MicroRNAs (miRNAs), as a family of non-coding small RNAs, play important roles in the post-transcriptional regulation of gene expression. Sea cucumber (Apostichopus japonicus) is an important economic species which is widely cultured in East Asia. The longitudinal muscle (LTM) and respiratory tree (RPT) are two important tissues in sea cucumber, playing important roles such as respiration and m...
